Not sure about "one strike, instant DEATH", but...
I'm not sure about the DEATH portion of things, but the when Japan was nationalizing Karate shortly after WWII the All Japan Karate Federation's motto (or maybe it was an informal one in thier publications) was something like "Karate: one strike - certain defeat". I'll have to do a little more digging on this at home, but I think this is where Hollywood may have gotten the idea.

Actually... The abductors are the "bad girl" muscles. They open up the nutcracker. That is what I tell my daughter and other untrained young people if they are attacked and can attack back.
Go for the following:
-Eyes to interfere with their vision....they can't get to you well if they can't see.
-Throat to interfere with breathing....gagging or gasping for breath will slow them down so you can follow up or run.
-Knees to disable them from chasing you....any object to take out the knees or kick to the side with blade, or sweep with foot or object.
Also Bill is a big proponent of attacking to areas that induce a reflex reaction like the inner crease in the thigh/leg, back of knee, inside elbow, etc.
All good stuff.

I got a good trivia question..
Anyone remember the name of the movie members of the Mattson Academy were in, filmed in NY sometime in the late 60s, early 70s?
I was invited to bring a team from the academy by the director.
The film "stars":
G. Mattson, Jim Maloney, Bob Campbell and I can't remember the others.
We filmed a competition sparring match.
There was a Korean, fresh from Korea there, attempting to break a bunch of items. Everything broke as predicted except the large beer bottle, which he attempted (repeatedly) to break over his head.
After the third try, he began bleeding profusely, but he wouldn't hear of "giving up". On the four try he swung the bottle with all his might. A resounding Claaaang could be heard throughout the studio. It was his last attempt. Staggering, apparently "out" on his feet, he finally waved the cameras off as he went to a corner and fell over.
I remember viewing the film with a group from the academy. It was playing in a seedy "combat zone" theater. I was told it only lasted a week!
I would really like to see that video, especially the bottle breaking scene!

My understanding of the "one strike, instant death" (or "one strike, one kill") is that when mentioned it is promoted as a goal we should strive for, rather than either an automatic fact or a myth. It has happened in real situations, most likely by chance and shear crushing strength rather than technique, but it would be a good skill to have.
I've also heard this phrase used to describe the differences between Japanese and Chinese hitting philosophies. Japanese karate adopts the "one strike one kill" from kenjutsu, iaido, and samurai strategy. Chinese quanfa tends to promote overwhelming your opponent with a flurry of blows, each of which would not necessarily be debilitating by itself. Which of these strategies Okinawan karate uses tends to depend on who you ask.

Mike, how are you triggering their gag reflex?
If someone is within the range of their usually smaller foe, they can grab, and it'd be a challenge to land a blow that would deprive them of sight and wiggle out, esp since ducking and blinking alone make it hard to get in there. Unless you've got a tool. The throat is pretty easy to protect too--the sternal notch is always going to be vulnerable but actually crushing a windpipe is a major task.
While there are good KOs in a clench I don't see these as either likely kills OR as the one strike envisioned by most of whom that have talked about this to me. They always have a "grapplers will be unconscious before they get anywhere near me" attitude which I think was duly challenged in the UFC series. Meanwhile the goal of getting to a perfect shot, even if its a goal and not an illusion of certainty, illustrates for me the old surgical motto,
"Perfect is the enemy of good."
People are going to miss chances to overwhelm someone with a series because they think they're going to drill someone with their hammer of thor.

You should try to contact the original company involved or previous owners.
I remember my first sensei Bob Blaisdell showing us a commercial which aired during the superbowl in the mid to late seventies.
I can't recall if Mike Murphy was on it or not.

Ian, I'll dig up my Gray's Anatomy so I can tell you exactly where, but some are glands underneath the jaw and others on the side of the neck by the sternocleidomastoid muscle (LI 21). I've had it done lightly on me and it got me gagging and coughing, and that made it hard to stay in the fight. A guy I know used it on a drunk who was smacking around his girlfriend and it supposedly got very colorful. Applied more gently you can use it as part of some nice controls. Nothing magic, just another tool to put in the box that goes very well with the Uechi ones.Mike, how are you triggering their gag reflex?
Yeah training films, yeah that's right. Not that I ever saw any of those, uh training films.
